The best way to determine if a female guppy is pregnant is when her belly will get large and have a squarish appearance to it. All females show the black spot in the gravid area. It just gets darker before the female gives birth indicating there will be fry soon. Before a female gives birth she will isolate herself from the rest of the group. Typically she'll hide somewhere as close to the surface as she can get (i.e. behind the heater/filter) and won't usually give birth until late at night when everything is dark. If she is thin the next morning but you can't find any babies it is probably because the adults ate the babies. Guppies aren't exactly the parenting type.
